The film opens in January 2004, where a household in Wasseypur is watching a daily soap. Suddenly, the TV is blown up by a gunshot. Sultan Qureshi (Pankaj Tripathi) along with his henchmen are scouring the locality, killing suspicious passerbys on the way. They finally reach a sprawling house and open fire, riddling the house with bullets. Sultan then makes a call to a number,which starts ringing. It is revealed that he's calling Faizal Khan's(Nawazuddin Siddiqui)number and on getting no response, Sultan assumes that Faizal and his whole family have been killed in the gun fire and leaves, unaware that the family actually survived the gunfire and are safe in the house. Sultan then calls JP Singh, son of Ramadhir Singh (Tigmanshu Dhulia)to inform about the hit, when he notices a police checkpost on the road. He inquires JP about this development after which JP abruptly disconnects the call, confirming suspicions that the checkpost is for eliminating Sultan. A crossfire ensues leaving Sultan hit.
As the opening credits roll, the narrator gives a whole history of Wasseypur, a city in Dhanbad predominant with Muslim inhabitants. The town is divided into 2 sections- The powerful Qureshis, butchers by profession, and the other muslims. In 1941,during british regime, Sultana Dacoit,a Qureshi, used to rob goods and precious items from the British trains coming into the town. Shahid Khan (Jaideep Ahlawat), taking advantage of the fact that nobody has seen Sultana's face, robs food grains and sells it in the town. After Sultana finds out, Shahid along with his pregnant wife and friend Farhan (Piyush Mishra) are banished from the town and they settle near dhanbad where Shahid works in the coalmine. During childbirth, his wife dies. Shahid is enraged as he wasn't with his wife because of the coalmine operator who didn't allow him to leave on time. In a fit of rage he kills him. After independence in 1947, the coalmine is bought by Ramadhir Singh from the British. He employs Shahid Khan as an enforcer in the coal mine & becomes his trusted man. However, Ramadhir eavesdrops on Shahid's & Farhan's conversation where the former has plans to rule the coal mine one day. A furious Ramadhir leaves, leaving his umbrella near the main door. Ramadhir sends Shahid to Varanasi to get some new weapons from a dealer, who eventually kills Shahid. Farhan notices Ramadhir's umbrella near the door, concluding that Ramadhir eavedropped on their conversation. He flees from the place with Shahid's son, Sardar in the nick of time as Ehsaan Qureshi (Vipin Sharma) a henchman shows up. Ehsaan lies to Ramadhir that he killed Farhan & Sardar. After hearing of his father's death, Sardar shaves his head vowing that he won't grow his hair until Ramadhir is dead.
20 years later, Sardar Khan (Manoj Bajpai) along with his cousin Asghar (Jameel Khan) start a transport business ,whereas Ramadhir Singh becomes a well known politician,and singlehandedly runs the coal mafia with an iron fist. Sardar gets married to Nagma (Richa Chadda), and eventually his son Danish is born. With revenge on his mind, Sardar illegally usurps Ramadhir's land leading to their first meeting which ends in a scuffle between Sardar ,Asghar and JP. Sardar & Asghar are sent to jail for hitting JP, a politician. Ramadhir chides Ehsaan for lying to him about Sardar's death. Sardar and Asghar learn how to make bomb and escape from the jail and take shelter at an associate's place where they meet Durga (Reema Sen) a Bengali who takes a fancy to Sardar & vice versa. Sardar gets into a physical relationship with Durga, leaving Nagma and the 2 kids Danish & Faizal heartbroken.
In the 80's, Sardar becomes one of the feared men in Wasseypur, where he has to contend with Sultan & the other Qureshi's and in Dhanbad, where Ramadhir is his enemy. After a fight, Sardar leaves Nagma & goes to Durga leaving the former devastated. Faizal witnesses his mother in an amorous condition with Farhan and bolts out of the house. This incident pushes him to weed addiction and lawless activities. Sensing Sardar's increasing clout,Ramadhir employs Sultan (Sultana dacoit's nephew) as a henchman to counter Sardar, at the same time retaining his Muslim votes.
In 1990, coal mining becomes redundant and other activities like iron mongering rises as a route to making money. Sardar's son, Danish joins him in this business who in turn pleads his father to come back home. In an encounter with Sultan, Danish gets injured, an incident which leads to Sardar & Nagma reconciling. Durga doesn't approve of Sardar's proposal of living with Nagma together & tells him to leave her and her son alone. Sardar sends his second son Faizal to get some new weapons but gets caught by the police with the weapons in the train and is jailed. After sometime, Sardar renounces all lawless activities and starts a fishery business as a legitimate business. Meanwhile, Danish falls in love with Shama Parveen (Anurita Jha), who turns out to be Sultan's cousin. Eventually they marry, enraging Sultan. Faizal also is released from jail and starts a romantic relationship with Mohsina (Huma Qureshi (actress)). Faisal takes revenge from the dealer who sold him the guns by killing him, mirroring a scene earlier where the same arms dealer had killed his grandfather Shahid Khan on Ramadhir Singh's orders. After a night of merriment, Faizal inadvertently lets it slip to his friend that his father would be going out alone & unarmed the next day. While Faizal is sleeping, his friend informs Sultan about this piece of information.
In the climax, Sardar leaves home alone.After meeting Durga & giving her some money, he stops at a petrol pump to fill his car. He sees Sultan and his associates who come from behind in bikes and open fire on his car & escape. The film ends with Sardar staggering out of the car ,with multiple bullet wounds, and ultimately collapsing to his death.
